ABSTRACT

EVALUATION OF ANTICATARACT ACTIVITY OF SCAEVOLA TACCADA ON GLUCOSE-INDUCED CATARACT IN GOAT LENS: AN EX-VIVO APPROACH

Background: Cataract are a common eye condition that affects many people, particularly as they get older. They occur when the lens of the eye becomes cloudy or opaque, which can cause blurry vision. The development of cataracts is associated with oxidative damage to the lens proteins, which leads to protein aggregation and clouding of the lens. This research has been carried out to evaluate the anti-cataract potential of Scaevola taccada by using glucose induced goat eye cataract model. Methods: Goat eye lenses separated to five groups were incubated in synthetic aqueous humour culture, added by streptomycin of 0.1 mg/ml at 37°C for 72 hrs. Group I use as the normal control. Group II (disease control) the lenses were added by 1 M glucose. The lenses in Groups III (standard control) incubated in aqueous humour medium with 1 M glucose added by 500µg/ml of gallic acid. The lenses in Groups IV (Test Group) incubated in aqueous humour medium with 1 M glucose added by 500µg/ml of extract of Scaevola taccada. MDL levels and catalase activity were measured by using UV Spectroscopy. Statistical analysis parameters was conducted using ANOVA and post hoc Tuckey test. Result: As a part of study, In MDA level estimation, our test drug has low MDA level as compared to the disease control. So, it may be acting as anti-cataract. In total protein estimation, test drug has higher amount of total protein in comparison with others. By analysing catalase activity, Catalase activity data revealed that test drug is having high catalase activity as compared to standard control group. So, we conclude that it works as anti-cataract.    Conclusion: The study carried out revealed that Scaevola Taccada (Bhadrax) has potential for anti-cataract activity. This hypothesis was supported by finding total protein level, MDA level, catalase activity.
