SOLUBILITY ENHANCEMENT TECHNIQUE OF CINNARIZINE BY MIXED HYDROTROPY

Cinnarizine complex is prepared by Mixed Hydrotropy method by using four different hydrotropes such as Sodium citrate, Sodium acetate, Resorcinol and Niacinamide. The prepared Cinnarizine complex were then subjected for characterization of FTIR, DSC, SEM and XRD, solubility, drug content, percentage yield, Micromeretic property and in-vitro drug dissolution. The solubility and dissolution rate of prepared complexes were significantly higher than the pure drug. Initially the solubility of drug is determined by different concentrations of individual hydrotropes viz 10, 20, 30 and 40. The highest concentration of hydrotropes gives the highest solubility enhancement ratio. Drug content and percentage yield shows highest in F10 and F13 formulations. The FTIR and DSC of the prepared hydrotropes indicate that the drug and excipients are compatible with each other. The surface morphology of the prepared hydrotropes shows smaller crystals having very sharp needle in shape with crystalline surface compared to pure drug. XRD of prepared hydrotropes shows increases intensity of peaks compared to pure drug. Micromeretic property of solid dispersions shows good flow property. The F10 formulation shows highest drug release of 84.72±0.359 and it follows first order kinetics with non-Fickian drug transport mechanism. The Cinnarizine hydrotropes was formulated successfully using combination of lower concentrations of hydrotropic agents to increase the solubility and dissolution.
