Synthesis and SAR of Isoform selective HDAC6 inhibitors of novel imidazoquinazolinones is shown. It demonstrates HDAC6 selective pharmacokinetic properties and has clean off-target profile. It shows efficacy comparable to approved HDAC inhibitors.
